Synopsis "Développement et validation de méthodes pour l'estimation du voriconazole (in French)"

Une méthode RP-HPLC rapide, sensible et spécifique avec détection UV a été développée et validée pour la détermination du voriconazole. La chromatographie a été réalisée sur une colonne Enable C18 G (250 × 4,6 mm) en mode isocratique, en utilisant une phase mobile de méthanol et d'acétonitrile (60:40, v/v). Le débit a été fixé à 1 mL/min et l'effluent a été contrôlé à 223 nm. L'analyse chromatographique a révélé un pic principal pour le voriconazole à un temps de rétention de 2,841 minutes. Le méthanol a été choisi comme solvant pour l'analyse, et le λmax a été déterminé à 223 nm. La méthode a montré une linéarité sur une plage de concentration de 5-30 g/mL. Les paramètres de validation, y compris la linéarité, la gamme, la précision, l'exactitude, la limite de détection (LOD) et la limite de quantification (LOQ), ont été évalués et ont satisfait aux normes requises.
