Synopsis "Rekombination (in German)"

Zellpenetrierende Peptide (CPP) sind hauptsächlich kationische Peptide, die in der Regel aus einer Zusammensetzung von 5 bis 30 Aminosäuren bestehen und in der Lage sind, die Zellmembran zu durchdringen. Durch die Fusion von Molekülen, die selbst nicht in der Lage sind, die Zellmembran zu durchdringen, mit dem CPP wird eine Infiltration möglich. Das Genkonstrukt (kodierend für TS1-218+CPP) wurde aus der gereinigten Plasmid-DNA (pET 26 b (+)) herausgeschnitten und mit dem Vektor pPICZαA verbunden. Diese rekombinierte DNA wurde in E. coli elektroporiert und in weiteren Schritten auf P. pastoris übertragen. Für zukünftige Studien wird es interessant sein, den Vektor mit dem Gen erfolgreich in die Pichia-Zellen zu transformieren. Darüber hinaus besteht das Hauptinteresse darin, ihre Proteinproduktion zu steigern und deren Wirkung auf epitheliale Krebszellen zu analysieren.
